Super-Resolution Imaging

Application

Fluorescence imaging of fixed and live samples at spatiotemporal resolution beyond confocal and Airyscan systems. 3D PALM/STORM localises fluorescent labels with ~10 nm precision and instant SIM (iSIM) images at ~150 nm resolution at speeds upto ~170 frames per second. The iSIM also provides rapid acquisition of thicker volumes at this resolution.

The PALM/STORM is equipped with 405, 488, 532, 561 and 647 nm lasers and a x60 NA 1.2 objective for down to 20 nm precision in fixed samples. The iSIM is equipped with 488 and 561 lasers for two instant two-colour imaging of fixed and live cells with down to 150 nm resolution.

Equipment

  • 3D PALM/STORM: Fluorescence localisation microscopy (3D PALM/STORM) for ~10 nm precision of labels (fluorescent protein tag or dye-labelled antibody/other binder) in fixed samples. 405, 488, 532, 561 and 647 nm excitation lasers, 60x 1.2NA water-immersion objective.
  • iSIM: Instant, two-colour, structured illumination fluorescence microscopy with ~150 nm resolution, allows real time viewing of fluorescence images. Video capture at frame rates up to ~170 nm frames per second (fps) for live cells. Rapid Z-stack and/or time lapse imaging capabilities. 488 and 561 nm excitation lasers. Objectives: 60x 1.2NA water, 60x 1.3NA silicone oil and 60x 1.4NA oil-immersion lenses.
